How do I insert a significance symbol over a plot(bar chart)?

I am not sure if you mean an alpha or a sigma, but both are available in the Character Map.
Add a text object to your graph
Press Ctrl+M to open the Character map
Select the character, click Insert then click Close

You can also type the english equivalent (a or s?) then select it and click the Greek toolbar button.
