I am trying to integrate peaks from raw data. I'm using Origin 6.0. The Pick Peaks tool successfully finds and labels the three positive and one negative peak. After selecting a data range which excludes the negative peak, I tried using the Baseline Tool with mixed success. The baseline is created, modified and subtracted unsuccessfully. The Peaks Tab results in only one peak, the highest, being identified, yet the integral displayed from the Area Tab appears to be correct for all the peaks. Again, only one set of results, the highest peak is written to the Results Log.(I have successfully integrated the next smallest
peak if I narrow the data range to surround it.) Do I have to integrate each
peak separately to have results tabulated?

You would probably get more help by contacting Technical Support directly.

You do not mention how you excluded the negative peak and it's not clear from your description how the subtraction is 'unsuccessful'.
There are also a number of options in the tool that you don't mention that have significant bearing on your results.
Here are some things to consider:

  • When you subtract a dataset, any excluded range of data will be excluded from the subtraction.
  • Once you have subtracted a baseline, there is no difference between integration from baseline or integration from zero (because subtraction 'zeroes' the baseline).
  • Integration will give results for an entire dataset unless you have checked Use Base Markers.
  • If Use Base Markers is checked AND no base markers were set between any two peaks (those peaks overlapped), THEN the area for both overlapped peaks will be reported as the same in Results.

You can do integration analsis on a peak-by-peak basis (using the Data Selector), but if the Peaks Tab on the Baseline Tool is finding good Base Markers for each peak, then integrating with Use Base Markers checked will report areas for each individual peak in one step.
