Posted - 01/30/2017 : 07:42:21 AM I have recently acquired fluorescence data that I wish to use to plot the emission data (the broad baseline). However, the data also shows sharp peaks attribute to another process (Raman scattering) that I wish to remove. Ideally I would be able to take my data set, find/fit the baseline.
I have tried using the standard baseline finder in peak analyser; however, the output I obtain requires a lot of manual additions and deletions. Even after this the resultant baseline (fluorescence signal) is not very smooth and I find a significant variation of the peak emission value (peak of the broad baseline) after several repeat attempts.
An example of the type of data I am working with can be found here (google drive): https://drive.google.com/open?id=0B4YTJ3V-IqLfamVaMmxuZ0xIWVk
Any help/suggestions to resolve this problem would be greatly appreciated.
Thanks!

How about applying the median filter as in this document? http://www.originlab.com/doc/Origin-Help/Smoothing
Hope this helps.
