 with Origin?
 
 e.g.
 
 2
 2
 5
 5
 6
 
 All 2s should get 20 and all 5s changed to 50.

 1) Highlight the column, and select Column:Set Column Values menu.
 2) Enter the following formula with the conditional operators like:
 col(A)[i]==2?20:(col(A)[i]==5?50:col(A)[i])
 
 There should be many ways to achieve that.
