dear all,
I am looking for a way to get the mean grey value in a box inserted in an open picture with origin as in the picture below.

Hi Bedibra,

Please crop the region out by using the Crop tool,

http://www.originlab.com/doc/Origin-Help/Image-Crop

And, then, run 
mstats

in Command Window or Script Window to get the mean value.
