I have two 8-bit grayscale 6400dpi images as tiff files [I would like to keep their resolutions highest possible throughout the analysis process in Origin]. One image is the background/noise; another is the data/signal. I can convert both to matrices, but am not sure how to manipulate them. e.g., I would like to subtract the matrix of the 1st image from the 2nd one, to give me the actual signal. However, as both images are generated from a scanner, and do not have the exact same positioning with respect to the objects I scanned, the matrix elements differ by some horizontal & vertical offsets...could I somehow overlay one image over another, move them relative to each other, and then choose the region of interested to be subtracted?

Another question: If my scanned object happens to be slanted, could I somehow skew/'horizontalize' it, so that the straight edge of the object is parallel to the image border?

Hi Amy,

If you have Origin 7.0 or higher you can use Origin C to extract submatrices (possibly selected by a region of interest tool). Otherwise you'll need to crop by deleting rows or columns from the edges. You can rotate matrices by 90 degrees in both LT and OC but that won't help with your skew.

It might be easiest to massage the images in a good graphics program (if you have one) before importing them to Origin. Paint Shop Pro, for example, will rotate an image by an arbitrary angle and most graphics programs will crop an image.
