Origin Ver. 2024b and Service Release (Select Help-->About Origin):
Operating System: Windows 11

Hi, I am finding it difficult to recreate the plot attached. Basically I want to plot the curve and then assign colours to intervals e.g. 0 - 20 yellow, 20-100 blue. The colours however are not associated with the data , it is just to differentiate between sections

You can add reference lines that span to the whole layer height and fill areas between reference lines. see https://www.originlab.com/videos/details.aspx?id=170

You don't have to create a column D for the intervals. You can directly enter 0 20 100 ... on the top of the Reference tab to set the intervals
